Preservation Methods, Graft Storage, and Rehydration Guide

Body
Preservation Method About the Method Storage Temperature Suggested Re hydration Time
Fresh
  • The technique is dependent on minimal preparation and rapid cooling of freshly obtained tissues
  • This method can maintain viable cells until implantation
  • Refrigerated 1-10°C
  • NA
Frozen
  • Allografts are cleaned, disinfected, and may be sterilized prior to freezing
  • Long shelf life
  • Frozen -40 to 80°C
  • Room temperature solution soak: 5 minutes
  • Warm solution soak: 3 minutes
  • Room temperature (standing air): 20 minutes
Cryopreserved
  • Graft contains cryoprotectant and are frozen at a controlled rate, both of which contribute to preserving cell viability upon use.
  • Maintains biomechanical properties
  • Long shelf life
  • Liquid Nitrogen -120°C or below (for cardiac grafts)
  • Or Frozen -40 to 80°C
  • Room temperature standing air thaw for 2 minutes (Cardiac grafts for 7 minutes)
  • Followed by a warm sterile saline solution soak (minimum 2000ml and maintained at 37°C-42°C).
Freeze-Dried
  • Also known as lyophilization
  • Tissue preserved by this method requires rehydration of the grafts
  • Easy Storage
  • Long shelf life
  • Ambient Temperature
  • Rehydrate until required consistency and handling are achieved as per physician preference.
  • Keep graft fully submerged until implantation
Glycerol Solution(preservon*)
  • Preservon is a proprietary, glycerol-based preservation solution that allows allograft bio-implants to be stored in a fully hydrated state at a ambient temperature. This eliminates the need to freeze or freeze-dry allograft bio-implants.
  • Easy Storage
  • Long shelf life
  • More interoperating room options by saving valuable operating time
  • Maintains bio mechanical properties
  • Ambient temperature
  • Rinsing is not required but may improve handling
  • Per physician preference, the bio-implant can be rinsed in sterile irrigant prior to implantation
